Methanol Extraction:

 

Methanol Extraction Procedure:  Methanol extraction method is useful for characterization at the phylum-level.  Methanol extraction method only removes the naked pigment.

In vivo Bead-Beating:

 

In vivo Bead-Beating Procedure:  In vivo extraction removes the pigment with the protein it’s bound to.  It’s useful for characterization up to the genus-level.

DISCUSSION:

Green sulfur bacteria presence is indicated by peaks at 750nm in the in vivo, and 667nm in the methanol. 

Purple/ proteobacteria presence is indicated by peaks at 795nm and 890 nm in the in vivo, and 769nm in the methanol.
